Origin of Method (Published or unpublished reference):
Unpublished originated 1977 Dr.Ed. Lane, A & Laboratiriers, Omaha, Nebraska.
Reagents (and sources where applicable)
- 99ml sterile D.I. water blanks.
- 99 ml sterile buffer dilution blanks consisting of 1.25 ml stock phosphate buffer and 5ml of stock Mg SO4 per liter D.I water Sterilize at 121oC for 15 minutes.
- Media, MRS Lactobacillus agar, Difco labs.(See appendix 1 when removing media from autoclave, swirl very gently.
Apparatus (and sources where applicable)
- Rotary shaker or wrist action shaker
- Colony counter
- General laboratory glassware including screw – top bottles for dilution blanks
- Blender, commercial type similar to Oster brands
Standard Solutions:
- Stock Phosphate Buffer – 34g.KH2PO4 per liter D.I. water with PH adjusted to 7.2 using IN NaOH
- Stock MgSO4 Buffer 50g MgSO4-7H2O per liter D.I water
Procedure:
- Weigh 1 gram of sample and place in a sterile D.I. water blank. This is the 10-2 Dilution .
- Shake 18 hours . This is a necessary step as the lactobacillus bacteria are released and re-activated through this shake .
- Blend for 1 minute on the “Mix” setting
- Prepare standard 100 fold dilutions from the 10-2 dilution bottle through 10-8 in the 99 ml sterile buffer solution blanks.
- Plate 1.0 ml from all dilutions plus an additional 0.1ml from the 10-8 dilution creating a final dilution series from 10-2 through 10-9 (see Figure1)
- Pour 10ml MRS Lactobacillus agar at approximately 45° The agar is prepared per manufacturer’s instructions and sterilized for 15 minutes at 121°C. Allow plates to solidify at room temperature.
- Incubate inverted plates at 40°C for 48 hours AEROBICALLY .The Star Labs Lactobacillus bacteria is microaerophilic. (Autoclaving drives most of the oxygen out of the agar) 100% anaerobic conditions actually hinders bacterial growth and will lead to lowered counts.
- Count all subsurface colonies.
- Confirmation of lactobacillus sp. Bacteria is by gram stain. Lactobacilli are rod shaped, gram positive, catalase negative bacteria.
Note: Streptococcus faecium may be extracted using this procedure but should be counted on KF Streptococcus Agar (See Appendix 2)
It has come to my attention that there are potential difficulties in counting BIOPROL in blended or pelleted feeds. The difficulty lies in the presence of environmental Lactic acid bacteria and other naturally occurring bacteria in feeds which interfere with the counts.
The basic procedure should be amended as follows:
- An oven is required to maintain 45-50°C. A 25g portion of the sample is placed in a plastic bag and allowed to heat for 2 hours. After 2 hours the sample is placed in the 99ml water blank and extracted as normal. This should kill all vegetative cells. Leaving the stabilized BIOPROL culture.
- Proceed with the SL01 procedure by weighing a 1.0.g. sample.
- After the plates are counted, at random choose several colonies and gram stain them looking for gram positive bacilli and/ or gram positive coccobacilli. Also cheek catalase activity with 3% hydrogen peroxide Lactobacilli are catalase Adjust plate counts accordingly.
Note. Time and Temperature are related as far as this assay is concerned.
High temperatures require les time and cooler temperatures require more time.We have found the 48-50°C temperature for 2 hour to work best.Any adjustments in time or temperature should be compared to the suggested temperatures and time to determine the validity of the changes.

Special advice or precautions: When removing media fro, autoclave, swirl very gently. DO NOT SHAKE VIOLENTLY, as too much oxygen could be infused into the media for successful counting. The use of APPROVED MRS Lactobacilli Agars is essential for proper counting of these products. See Appendix 1.
It has been noted that differences in the equipment used to shake the culture (rotary or wrist action shakers) , as well as experimental trials using no shaker, can result in up to 100 fold differences in reported results. This is a mechanical problem with the procedure, dealing with the fracturing of bacteria in the culture during the extraction. Since it is unreasonable to dictate a specific shaker to be used, we recommend adjusting the shaker speed to a level vigorous enough to fracture these chains, in an attempt to eliminate these differences.
Appendix 1 This medium is sterilized by autoclaving for 15 minutes at 121°C A broth preparation is available through Difco laboratories, Detroit, Michigan. Bacto Agar (Difco) is added at the rate of 15g per liter to complete the media with sterilization as above . |
Appendix 2 Add 764 g agar per 100ml distilled water and sterilize for 10 minutes at 121°C. Cool to 45°C and all 1 ml of a 1d% solution of Triphenyl tetrazolium Chloride (TTC) |

Sensitive : Means the antibiotic will kill or destroy the organism (s)
Moderately Sensitive : Means the antibiotic may not destroy the organism(s) But it will help to a certain degree as far as Slowing the infection down.
Resistant : Means the antibiotic will not have any effect on the organism(s)
and should not be used to right the infection caused by the organism.
